Isolated chloroplast membranes of the unicellular photosynthetic alga Chlamydomonas reinhardtii were fixed, sectioned, and examined by transmission electron microscopy. Extensive regions of stacked thylakoids are seen in these wildtype cells. The section also includes the remnants of a mitochondrion (mi). See Fig 2 in Goodenough and Staehelin 1971 J Cell Biol 48:595-619.
Isolated chloroplast membranes were fixed with glutaraldehyde, post fixed with osmium tetroxide, dehydrated, embedded in epoxy resin, and thin sections cut, stained and examined with a Hitachi HU-11C TEM.
