In the case of a developing organelle, once the vesicles have grown to 1-2µm in diameter an electron-opaque crystal begins to grow at the center of the developing trichocyst. The paracrystalline material is finely striated. TEM taken on 12/9/70 by R. Allen with Hitachi HU11A operating at 75kV. Neg. 28,599X. Bar = 0.2µm.
Standard glutaraldehyde fixation followed by osmium tetroxide, dehydrated in alcohol and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness. The negative was printed to paper and the image was scanned to Photoshop. This digitized image is available for qualitative analysis.
