Xenopus egg extracts were mixed with sperm chromatin, treated to induce spindle formation, and antibodies added to deplete nucleoporin components. Rhodamine-tubulin was added, the preparations fixed with paraformaldehyde, and spindles centrifuged onto cover slips through a glycerol cushion.
