Description

Distribution of Bib1, BubR1 and Mad2(red) on chromosomes assembled from sperm chromatin in nocodazole treated Xenopus oocyte extracts in vitro. Chromosomes are stained with DAPI (blue). Right panels show the effect of depletion of the Nup107-160 nucleoporin complex. See Orjalo et al 2006. Mol Biol Cell 17:3806-3808.

Technical Details

Xenopus egg extracts were mixed with sperm chromatin, treated to induce spindle formation, antibodies added, and the preparations fixed with paraformaldehyde, and centrifuged onto cover slips through a glycerol cushion, processed for immunofluorescence, and examined using a Zeiss Axioskop microscope. See Fig 5A in Orjalo et al. 2006 Mol Biol Cell 17:3806-3818.
